Mustafa was born to Sindhi actor Salahuddin Tunio and studied to be a pharmacist before he quit to pursue acting. Mustafa had made his film debut in 2014 in Nabeel Qureshi's film 'Na Maloom Afraad'. Mustafa married Sana Fahad in 2006, who is also an actress and writer. The couple has a daughter named Fatima, and a son named Moosa.
