Chasing a target of 164, Mumbai Indians were bowled out for their second lowest total this season in just 12.5 overs.
Despite the loss, Mumbai Indians stayed on top of the points table with 16 from 11 matches while Kings XI are ninth with eight points from 10 matches
The famed Mumbai Indians' batting struggled against the spin of 21-year-old left-arm spinner Bharghav Bhatt, who finished with figures of four for 22 and was also adjudged the Man of the Match.
Only four Mumbai Indians batsmen - Kieron Pollard (17), Aiden Blizzard (15), Ambati Rayudu (13) and Harbhajan Singh (12) - could get to double digits.
Medium-pacer Praveen Kumar struck in his successive overs to get rid of Sachin Tendulkar (6) and Blizzard, while a freak run out of Andrew Symonds (8) triggered a collapse that saw Mumbai Indians losing seven wickets for just 43 runs.
Earlier, a five-wicket haul by Munaf Patel (5-21) restricted Kings XI to 163 for eight in 20 overs.
Shaun Marsh was the top scorer for the hosts with 43, while Dinesh Karthik made 31. Skipper Adam Gilchrist made a run-a-ball 28.
