According to the police, the incident occurred late Saturday night when a speeding Maruti van, on its way from Haryana to Agra, collided with a matador coming from the opposite direction.
The bridge, which was undergoing repair, was partially open for two-way traffic.
Five people died on the spot and another person died in Mathura's Swarna Jayanti Hospital, they said, adding that the condition of the four injured was critical.
Superintendent of Police (city) Ram Lal Verma said the victims had not yet been identified but they appeared to be 25-40 years old.
