The big-serving South African smashed 15 aces and 81 winners to claim a 7-6 (7-5), 6-3, 6-7 (7-2), 7-6 (7-0) win over Murray in a gruelling contest that lasted four hours and 18 minutes at Flushing Meadows.
Reflecting on his victory, Anderson said that he just managed to keep his composure throughout, adding that he probably played one of the best matches of his life, the Guardian reported.
Murray, the 2012 US Open winner, squandered too many chances as he failed to convert eight of 11 break points.
Anderson will next face French Open champion Stanislas Wawrinka in the quarters.
